چکیده

Abstract Activation and proliferation of antigen specific T cells depends on the interaction multiple signaling molecules with TCR/CD3 complex. We report that expression connexin 43 (Cx43), a gap junction protein, is critical for expansion antigen-specific or self-reactive effector CD4 +T cells. While mediated intercellular communication has been main focus research, cytoplasmic C-terminus Cx43 functions independent membrane channel forming properties may act as docking molecule assembling complexes including immunological synapse. also Cx43-deficient transferred into recipient mice immunized cognate antigenic peptide adjuvant expanded poorly compared to wild type This protected from experimental autoimmune encephalomyelitis (EAE). Transcriptome RNA-seq analysis comparing gene profiles activated sufficient shows differentially expressed genes are enriched in involved G protein Ca 2+signaling, ATP metabolism, regulating cytoskeleton reorganization cell homing. In summary, our data demonstrate major impact generation their ability respond stimulation, precipitate

Gap junction protein connexin-43 interacts directly with microtubules

Gap junctions are specialized cell-cell junctions that mediate intercellular communication. They are composed of connexin proteins, which form transmembrane channels for small molecules [1, 2]. The C-terminal tail of connexin-43 (Cx43), the most widely expressed connexin member, has been implicated in the regulation of Cx43 channel gating by growth factors [3-5]. The Cx43 tail contains various ...

Polyamines regulate gap junction communication in connexin 43-expressing cells.

The control of cell-cell communication through gap junctions is thought to be crucial in normal tissue function and during various stages of tumorigenesis. However, few natural regulators of gap junctions have been found. We show here that increasing the activity of ornithine decarboxylase, or adding polyamines to the outside of cells, increases the level of gap junction communication between v...

Induced Gating of Connexin 43 Gap Junction Channels

Suppression of gap-junctional communication by various protein kinases, growth factors, and oncogenes frequently correlates with enhanced mitogenesis. The oncogene v -src appears to cause acute closure of gap junction channels. Tyr265 in the COOH-terminal tail of connexin 43 (Cx43) has been implicated as a potential target of v -src , although v -src action has also been associated with changes...

Myosin VI facilitates connexin 43 gap junction accretion

In this study, we demonstrate myosin VI enrichment at Cx43 (also known as GJA1)-containing gap junctions (GJs) in heart tissue, primary cardiomyocytes and cell culture models. In primary cardiac tissue and in fibroblasts from the myosin VI-null mouse as well as in tissue culture cells transfected with siRNA against myosin VI, we observe reduced GJ plaque size with a concomitant reduction in int...

Rescue of perfluorooctanesulfonate (PFOS)-mediated Sertoli cell injury by overexpression of gap junction protein connexin 43

Perfluorooctanesulfonate (PFOS) is an environmental toxicant used in developing countries, including China, as a stain repellent for clothing, carpets and draperies, but it has been banned in the U.S. and Canada since the late 2000s. PFOS perturbed the Sertoli cell tight junction (TJ)-permeability barrier, causing disruption of actin microfilaments in cell cytosol, perturbing the localization o...
